    Impact Statement: Our workflow allowed profiling of known and unreported Urinary mercapturic acids (MAs) from endogenous and environmental sources, and the levels of several MAs were increased in smokers. Our method can also be expanded and applied to other exposure-wide association studies.

Through analysis of diverse demographics and tobacco usage profiles, this dataset offers a distinct opportunity to comprehensively investigate subclinical and clinical CVD risks, including within underrepresented groups.

    Impact Statement:  We established a well-designed cell co-culture system to investigate the reciprocal interaction between human NCCs (hNCCs) and human PCs (hPCs), and also monitored the migration behavior of NCCs and PCs in zebrafish embryos. We found that ethanol exposure resulted in a disruption of coordinated hNCCs-hPCs interaction, as well as in zebrafish embryos. Treating hNCCs-hPCs with exosomes derived from ethanol-exposed hNCCs (ExoEtOH) mimicked ethanol-induced impairment of hNCCs-hPCs interaction. We also observed that SDF1, a chemoattractant, was downregulated in ethanol-treated hPCs and zebrafish embryos. Meanwhile, miR-126 level in ExoEtOH was significantly higher than that in control exosomes (ExoCon). We further validated that ExoEtOH-encapsulated miR-126 from hNCCs can be transferred to hPCs to suppress SDF1 expression in hPCs. Knockdown of SDF1 replicated ethanol-induced abnormalities either in vitro or in zebrafish embryos. On the contrary, overexpression of SDF1 or inhibiting miR-126 strongly rescued ethanol-induced impairment of hNCCs-hPCs interaction and developmental defects.

    Impact Statement: Fetal alcohol spectrum disorders (FASD) are the most prevalent nongenetic birth defect in Western countries and a leading known cause of intellectual disability. This review article provides an overview of the current knowledge on the detrimental effects of ethanol on neural crest cell (NCC) induction, migration, differentiation, and survival. It also explores the intricate molecular mechanisms underpinning these effects. The article also spotlights potential therapeutic strategies for preventing or mitigating the detrimental effects of ethanol on NCCs and reducing the risk of FASD. Overall, this article offers a comprehensive overview of the current understanding of the impact of ethanol on NCCs and its role in FASD, and highlights the remarkable contributions of Dr. Shao-yu Chen’s laboratory to FASD research over the past three decades.

    Impact Statement: Recent epidemiological studies reported significant associations between dietary heterocyclic amine (HCA) exposure and insulin resistance and type II diabetes, and our laboratory recently reported that HCAs induce insulin resistance and glucose production in human hepatocytes. The current study for the first time showed that insulin resistance and glucose production following HCA exposures in human hepatocytes is modified by the N-acetyltransferase 2 genetic polymorphism, reflective of precision environmental health.

    Impact Statement: The publication focused on understanding the longer-term impact (31 weeks) of PCB exposures on the liver and demonstrated that with longer duration of exposure, PCBs were able to induce more severe liver injury outcomes including liver cancer development. To our knowledge, this is the first study to demonstrate that PCBs, at doses relevant to current environmental exposure patterns, resulted in progressive liver disease outcomes, and underscores the importance of considering ‘duration of exposure’ when assessing hepatotoxicity indices for these chemicals.

Prior to the pandemic, patients in urban counties had significantly higher odds of opioid overdose death compared to rural counties, but during the pandemic, the distinction between urban and rural areas diminished, with opioid overdose mortality rates converging. The COVID-19 pandemic introduced challenges that exacerbated opioid mortality trends, affecting both urban and rural populations, reflecting the complex interplay between social, economic, and healthcare factors.
