Background

David J. Tollerud, M.D., M.P.H. is Professor and Chair of the Department Environmental and Occupational Health Sciences at the University of Louisville School of Public Health and Information Sciences. He received his M.D. degree from Mayo Medical School in 1978 and his M.P.H. degree from the Harvard School of Public Health in 1990. Dr. Tollerud has extensive clinical training, with specialty board certifications in internal medicine, pulmonary and critical care medicine, and environmental and occupational medicine, with a long-standing interest in the health of disadvantaged and underserved populations.

Dr. Tollerud's research interests include the effects of environmental pollution on asthma and other health problems, particularly among children and inner-city disadvantaged populations, nanoparticles, and stem-cell transplants. A second area of research interest is strategies to prevent work-related injury and illness. Dr. Tollerud has published over 60 scientific articles, books and book chapters, is a member of numerous professional and scientific organizations, and sits on a number of local, national and international committees dealing with environmental, occupational, and public health issues.

Ongoing funded research includes:

National Children's Study-Jefferson County Center. This initiative provides critical new information regarding environmental influences on children’s health and develop secondary and tertiary sampling units within Jefferson County over a 25-year period. It is the largest government-funded long-term study of environmental and genetic effects on children’s health to be conducted in the United States.

Center for Environmental Genomics and Integrative Biology. The goal of this grant is to support outstanding basic and translational investigations into the etiology of environmental disease and the development of new approaches to manage these conditions.  The Integrated Health Sciences Facility Core’s central goal is to provide Center investigators with services and resources that will facilitate the translation of basic research finding into clinical or public health applications.  The

Tolerance Induction for Tissue and Organ Transplants.  The goal of this project is to safely establish mixed chimerism and induce tolerance using facilitating cell (FC)-based therapeutic approaches and nonmyeloablative conditioning.

Community outreach and environmental education, particularly to African American and Hispanic communities, is also an important and growing focus of Dr. Tollerud and the Department's activities that include collaboration with other community and state agencies.

Completed research includes:

Health Effects of Occupational Exposures in PGDP Workers. The goal of this study was to carry out a feasibility study and pilot study to evaluate the health impact of occupational exposures on workers at the Paducah, KY Gaseous Diffusion Plant (PGDP) to determine if workers have a mortality rate which exceeds that of the general population, particularly for cancer. This study is an important contribution to our knowledge about the health of a key occupational cohort of workers that contain 6859 individual names from 1953-2003.

Stem Cell Graft Engineering to Treat Sickle Cell Disease (Phase II).  The goal of this project is to develop an approach to engineer a bone marrow graft to avoid the major complications of HSCT:  graft-versus-host reactivity (GVHD); graft failure; and the need for perfect matching.  Our approach is superior to current therapies because it involves a proprietary process that enriches for patented facilitating cells (FC) and stem cells but removes GVHD-producing cells.

Genetic and Environmental Risk Factors for PSP.  The goals of this study are to determine if there is an association between progressive supranuclear palsy (PSP) and the H1/H1 genotype, specific H1 sub-haplotypes, alpha-synuclein polymorphisms or parkin gene deficits, or if there are gene-gene interactions; if there is an association between PSP occupational and/or environmental chemical exposures functionally or structurally similar to rotenone/acetogenins; and if hypertension or traumatic brain injury prior to symptom-onset is associated with PSP. Understanding the etiology of PSP may help explain the causes of other tauopathies such as Alzheimer’s disease.
