Background:
The ProteinChip® System from Ciphergen Biosystems (Fremont,
CA) uses patented SELDI (Surface-Enhanced Laser Desorption/Ionization)
ProteinChip technology to rapidly perform the separation,
detection, and analysis of proteins at the femtomole quantities
using biological samples directly without the need for pre-analytical
extractions or sample-preparation procedures. The system allows
for analyte capture, purification, analysis, and processing
from complex biological mixtures directly on ProteinChip Array
surfaces. The signal amplification produced by the protein
capture and micropurification is analogous to searching for
“a needle in a haystack” by capturing the needles
and washing away the hay. Detection of the purified proteins
is performed by Laser Desorption / Ionization Time-of-Flight
mass analysis. Chemical and biochemical processing can be
included at any step throughout the SELDI process to greatly
enhance the detailed knowledge gained from a set of experiments.



Data analysis:
There are various ways of generating and representing data.

Data can be presented in a typical mass-spectrum. Several
patients or controls can be analyzed in mass-spectrometric
or gel-view formats:

Availability:
The SELDI facility is available in the Diagnostic Reference
Laboratory. Utilization by other researchers in our institution
will be made available by various collaborative arrangements.
